Abstract:
Substance use disorder (SUD) disrupts neuroplasticity and promotes neuroinflammation, potentially mediated by extracellular vesicles (EVs). EVs encapsulate molecular cargo reflective of their cells of origin, providing insight into SUD-induced brain changes. We investigated prefrontal cortex-derived EVs from postmortem human brains (Brodmann’s area 9, BA9) and rodent models of chronic morphine exposure, using multiomics profiling (proteomics, lipidomics, RNA-seq, Nanostring) to identify molecular alterations. Primary neuron cultures of mice were treated with brain-derived EVs (BDEs) to assess their impact on gene expression and glial activity.
